E’ da ormai un secolo che la razza umana è costretta a vivere senza la luce del sole. A oscurarlo fu Demitri Maximoff, potente vampiro che venne cacciato dal mondo delle tenebre e fece di quello degli esseri umani la sua dimora e il suo dominio. Dopo 100 anni, Demitri è pronto a dare l'assalto al mondo delle tenebre. Approfittando della situazione, gli umani iniziano a ribellarsi contro i DarkStalkers, le creature della notte, dando loro la caccia per sterminarli. Intanto Demitri incontrerà sulla sua strada la bella Morrigan, una nobile del mondo oscuro in cerca di un rivale degno della sua forza, e degli strani robot che attaccano sia gli umani che i DarkStalkers. En medio de una guerra entre las familias de Demitri Maximoff y Morrigan Aensland por el control del mundo demoníaco, el invasor alienígena Pyron llega a la Tierra y planea asumirlo sacando a aquellos que tienen la oportunidad de detenerlo, a saber, los Darkstalkers. Mientras tanto, el dhampir Donovan Baine trata de librarse de la maldita sangre que corre por sus venas. Cuatro de los Darkstalkers originales, Anakaris, Rikuo, Sasquatch y Victor no aparecen en la historia principal de la OVA, al parecer habiendo sido asesinado por Pyron en una secuencia de flashback mostrada al principio del cuarto episodio.

Vor 100 Jahren floh der Vampir Demitri Maximof aus der Dämonenwelt auf die Erde, die seitdem in einem Halbdunkel liegt. In dieser Situation haben sich auch viele andere »Darkstalker« auf der Erde niedergelassen, während die Menschen versuchen irgendwie zu überleben. Nun ist für Demitri die Zeit der Rückkehr gekommen und er stellt sich Morrigan, der jungen Königin der Dämonenwelt. Doch gerade als sie ihren Kampf beginnen, greifen geheimnisvolle Roboter an. Währenddessen versucht das Katzenmädchen Felicia sich in der Menschenwelt ihren Lebensunterhalt zu verdienen und gegen die menschlichen Vorurteile zu bestehen.
