衛子夫為漢武帝皇后38年，本劇參照《史記》、《漢書》相關記載，對漢武帝的第二任妻子衛子夫人生經歷進行描述，讓後人更為清楚的知道她是如何成為中國歷史上第一位有獨立諡號的皇后，也是在位時間第二長的皇后。劇情講述平陽公主府上出身低賤的歌伎衛子夫，因機緣巧合被漢武帝相中後帶入宮中，期間雖多次遭遇構陷仍化險為夷，並逐漸獲得漢武帝的寵愛，最終成為一代賢后。
