舞台は、徳川家康が豊臣政権を打ち破り、天下の覇者となった関ヶ原の戦い（1600年）から14年後の日本――。 豊臣方に加勢した武将・真田幸村は、高野山麓にある九度山に幽閉され、人知れず家臣として仕え続ける忍たちとともに、息を潜める生活を送っていた。 そんな中、若き豊臣家の当主・秀頼が反旗を翻すことを恐れた家康が、豊臣討伐を宣言したことから、事態は急変。 天才的な忍術を誇る忍者・猿飛佐助（岩本）と出会った幸村は、ついに時は満ちたと判断し、豊臣家を助けるため九度山からの脱出を決意！ 佐助は幸村への忠義を誓う忍たちと合流し、徳川家を相手取った最大の決戦＝大阪の陣へと突き進んでいくが…!?
